Liver DNA isolation

Yesterday (20 March 2009)
Task : Extraction the DNA from the liver tissue.

Before we start our experiment, our lecture is giving some briefing regarding some precautions and methods to take....


These are liver cells....


Before that, scalpel blade is 'install'...


Then the liver cell is minced into small pieces....


These are the minced liver cells...

Then, some of the minced liver tissues is transferred into sterile 1.5ml microcentrifuge tube... Lysis Buffer is added....
Followed by Proteinase K
Look!! I m seriously performed the experiments... Hehee...


After incubation for 1 hours, Phenol:Chloroform is added and it must be done in the Laminar flow hood....



Look!! These are the sample after adding phenol:chloroform (1:1)

Then, centrifuge the sample and u will obtain the results as below....
Transfer the supernatant (aqueous phase) into new sterile microcentrifuge tube...


Everyone of is busy perform their own experiments...


Look at the bottom of the tube.... There are the stringy DNA precipitate seen....


Hahah.... Look!! Our lecturer is busy telling his "story" to the lab demos and lab assistants....
