好消息！

我终于被Post 回来雪州了！

到底是好事还是坏事呢？？

不知道应该是高兴还是伤心是好呢？

高兴的：当然是回到自己最熟悉的地方。。。一切能够根据我的计划走，事业也许可以到达另一高峰。。。

（可是，就会忙到半死！)

伤心的：我是否有没有机会跟我心中的女神再续缘呢？

命运的安排，往往都在愚弄我。。。

（难道要我孤独终身嘛! :-)）haiz !!

焦虑！紧张！烦恼！

根据相关的情报显示我将在明天获知派往哪一个州属执教了！

到底我会在什么地方落脚呢？

雪州？吉隆坡？沙巴？砂拉越？

现在的心情一直在忐忑不安。

虽然一直口口声称到什么地方去都没有关系。。

但是就是不知道为何我不能淡定的等待结果呢？

现在心中都在焦虑着。。。

其实，我也很矛盾，不知道自己想去哪一个州属执教呢？

继续回旧校教书？是我一直期待的。。

可是，我现在改观了。。。留守在旧校也未必是件好事，

到时，我就会欠我校长一个非常的重大的人情。。

要怎么还都还不完。。（这时，我肯定会忙死了）

烦恼就多了，烦恼开始就会找上门来了！

如果，去到沙巴或砂拉越，也未免太远了吧！

我已经三年离家，到砂大读书，难道我又要被派去那里吗？

又是另外的三年？

沙巴的确是个不错的地方，至少有我追求的目标在哪里！

可是，又很担心，如果真的到了那里，我会去什么地方呢?

内陆？还是城市？

如果进了内陆，我的未来，我计划好的前途就毁了。。

我不想一生中满足于一个平平淡淡的老师。。

我想要更高的理想，我想当大学的讲师！

现在我还能做什么呢？也不是只是一个 “等” 字！

心啊心，能不能平静一下，好让我今晚有个美梦吧！

开学的第二天

2012 年1月5日

这一天，我有新的发现。。。
也成为学校老师们的焦点！再次当上了主角！哈哈！

（真不好意思，连续两天当上两天的主角）

我的第一个发现是，我发现今年一年级的新生来了一个过动儿。。。全体新生在礼堂集合时，他尽然一个到处拿着书包跑来跑去，跳来跳去，爬上台上自玩。。。

结果呢，训导老师只好把他带在她的身边，牵着他，
老师走到哪里，他就带着跟到哪里。。。

另外，我的另一个发现是我自己的班，
也有两个“比较特殊的”学生。。。就是 3A 班的。。

这两位学生就坐在我的前面。。。当我在进行交流时，其中一个学生就开始自言自语的，玩自己的东西，我尝试跟他讲话，他尽然不理会我，继续玩他的东西，讲他懂得语言。。。

（就是活在自己的世界里）

另一个学生呢，当我吩咐全班同学要准备几本作业时，
他就是傻傻的看着我。。。

我帮他拿了3 本簿子出来，要他在正当的空位里，
填上教师的名字及科目。。。

他还是一副傻傻的样子看着我。。。
我只好好好的指给他看，他才慢条斯理的抄下去。。

（如果换以前的我，我早就气爆了，一定会骂他的）

可是，进过师训后，态度完全变。。。
知道我以前所做的是错的。。。

我也参加过一个爱的教育讲座会，
从中得到很多不错的资讯。

现在我正在想我要如何教育这孩子呢？
我该如何尽我老师的责任呢？

真是烦恼着！我该如何做到两全其美呢？
不会耽误其它学生学习的时间，

同时也能这两位同学学到一些有用知识

天啊，请你给我一些指明吧！

到底什么事情我有当成了主角，成为全校的焦点呢？

那就是当天星期四，我穿上一件1 Malaysia 的Baju Batik 到学校。。。同事远远看到我走来，还真的把当成某某Datu... 教育部的高级官员哦！哈哈，开心死我了。。。

我被人当成Datu 哦。。。身份顿时升几级。。。
身价也涨了几倍哦！
（想不到一件RM 150 的衣服能够让我的身价翻倍）。

开学的第一天

2012 年1月4日是全国中小学的开课日。。。

正好这一天也是我第一天的上班日。。。

（这间华小并不是我开始合格执教生涯的第一所学校，而是我的旧校，因为此校严重缺少师资，一共少了5位教师，所以我就被校长及下午班主任叫回去暂时当上代课老师，以解决目前的燃眉之急）

那天，一如平时一样，准时到达学校。。。一踏进校门，我的感觉只能用一首诗来形容：

“ 年头离校年终回，校音无改鬓毛衰。

学生相见不相识，笑问师从何处来。 ”

只是，我的旧同事都很惊讶的我已经学成归来了，

每一个一见到我，都用即惊讶即兴奋的眼神来问候我。。。

" Eh ，金福，你终于回来咯？”

“金福，你被post 到这里来教书啊？恭喜你哦？”

“哇，你好快哦，这么快一年就毕业了？终于回来咯？来这里教书吗？”

而有些比较模糊的老师呢则问道，

“你现在是不是假期？暂时回来帮忙？几时还要回到受训呢？”

每个人左一句，右一句，我忙着不断回答他们的问题。。。

第一天上课嘛，所以主任就临时开了一个周会咯，让小一的学生认识校长啊。。。

要死不死的老师，回来的第一天都不放过我，一通电话就叫我上后台帮忙音响。

（虽然以前都是我是我负责的，可是你要知道我已经一年没有弄了啊！早就忘了一干二净咯）

结果，我还是帮不到什么忙，唱国歌及州歌时，CD 里的music 不能播。。。

无论我怎样乱按，还是播音机还是无法出声。。。结果只好叫我的朋友上来带唱。。。哈哈！

之后的校歌及Malaysia Berjaya 只好用出动我的电脑了。。。

世界名人之一———— 张帝

接下来想为大家介绍一位世界名人，大家都耳熟能详的，鼎鼎大名的张帝先生。。。
他堪称为乐坛上的奇才，能够随时现场现有的音乐，即兴填词做歌。。。因此，你享有急智歌王的美称！
他能也能称得上我们人类心灵的治疗师，凡是你有什么疑难杂症的说，可以找张帝。。

张帝本名是张志民，祖籍山东青岛市，是家中的长子。。

其中，他最著名的作品是1970年末代在Indonesia演出时创作的《毛毛歌》。

非常动听。你可以慢慢去聆听这首歌。

张帝，他呢，在还没有当歌手兼节目主持人前，是名外科医生。也许是受到他爸的薰陶下吧。在众多歌手当中，能读到医学研究生的人更是闻所未闻。不仅如此，张帝还曾被派到军队里当队医，成了一名出色的外科医生。

但是呢，张帝他本人是希望他自己的人生能够活得更精彩，更开心，与其每天面对着满面愁容的病患者，看着更多生死离别的场景，非常压抑的多日子，不如，做一些他喜欢做的东西，又能把快乐带给大家。最终，他还是选择放弃一个拥有显赫的社会地位和可观收入的医生，当歌手。

因此决定，不仅使张帝失去了饭碗，更让他被爸爸赶出家门。。

张帝，一路走来都是经历了不少的辛酸，忍羞受辱的局面。

记得一次，他在一家夜总会里会见老板，苦苦哀求老板让他在那里唱一两首歌，免费不收任何钱，可那老板拒绝了，说：位置已满了。

哪知，这时一位当时的红歌手从他们身边路过，老板飞速追上去，换个角色苦苦哀求那歌手来柱唱几首歌，多少钱都给得起。。。很显然的把一旁的张帝冷落。

这时的张帝，二话不说，静静地走到河堤边，落下了他的男儿泪。感叹社会上有如此现实。。

真的没有人会慧眼识英雄啊！

老天爷永远都回疼惜有才艺的人。。终于，让张帝摘到了一个机会，出头的一天了。

他在1972至1982，十年期间，参与了超过千场的演出，至今，这一纪录仍无人能够打破。

张帝曾金开创了华语歌坛的许多第一，比如第一个穿牛仔裤上台，第一个穿皮夹克上台，第一个留着大胡子

他曾讲过，

“原来人生是如此的可爱，

一个丑陋的外表，并不代表一个丑陋的人，

于是他告诉自己，

他愿意当人家心中的小丑”。

他非常的感谢老天爷给他恩赐，让能够做自己快乐的事，也能把欢喜带给大家。

每年的圣诞节的来临，就意识到我们要送别2011 年，准备迎接着2012年的到来。。。

今年也不例外。。。今年的圣诞呢，就比较孤单咯。。。

自己一个过。。也没有圣诞礼物。。

就在平安夜，陪着爸妈到Strait Quay 去凑热闹。。。

到了那里，爸妈俩就去“过着俩人世界”，唯有一个人四处走。。

累了，就坐在一旁，一边拿起手机，跟“某某人”讯息，一边享受圣诞佳节的音乐。。。

在我眼前经过的，全都是一对对的情侣，要不是就是一家人大小。。

咳。。唯我坐在独处。。:-(

冬至快乐！

今年跟往年不一样，今年过冬总算可以在家乡与家人一起过冬了。。。

昨晚，还帮爸妈做了一大蜗的汤圆呢。。。

萧煌奇的传奇

刚刚在网上（http://sugoideas.com) 观看一部最近在东风47 上映的王牌大明星。这集邀请来的明星有林志炫及萧煌奇。。这两位都是好朋友，而且对歌坛作出了伟大的贡献。。。

其中，另钦佩的是萧煌奇。。他是当今歌坛上，我认识唯一的盲人明星。。。因此对他那种坚韧不解的精神给佩服了。。很想知道他到底是什么原因的情况下造成他失明，使到他的世界顿时陷入的黑暗。。

在好奇心的驱使下，我就到GOOGLE 寻找他的个人资料。。

原来，他是一名视障歌手，出生后不久就患上白内障而失明。。但在4岁时，他成功了做了眼视手术，恢复了微弱的视力长达10 年。就在他15岁那年，因任性而过渡用眼，导致他完全失眠。。他曾经沮丧懊恼过，失意痛苦过，内敛的他，音乐陪他度过了最坚熬的日子。。

萧煌奇在 2002 年写了〈你是我的眼〉，并以这首歌卷全台，成为KTV 排行榜的常胜军。。他也写不少感人的华语及一些米南语歌。。。在他的职业生涯也荣纳了不少的奖项。。。他的精神是值得我们去学习的。。

Fall into the pond

Today i was reli bad luck.... I fall into small pond... Bruises my whole body including my sexy mouth... Haiz..

This afternoon, i finished my lab early... Around 4pm already finished lab work, so i went to BPPs (Bahagian Pendaftaran Prasiswarah) to obtain the examination remark form... Then on the way back to cempaka hostel, i pass by a fountain with a small pond near Center Language Studies (CLS)... Due to my laziness to go a big round, i decided to take shortcut.. I try to jump over the pond... But at last, i failed...

My right leg drop into the pond and fall down.. My right arm and shoulder hit the border of the pond... Then, my mouth direct kiss the floor... Bleeding alot...

So pain... Now, my mouth got abit bengkak liao... Haiz... Today reli shuai...

It was so embrassing.... : (

Meeting with Penang Vice Chief Minister

Reli not bad... New Government got new policy... Now the penang state government is so care about us who study oversea in East Malaysia... They wanna to be more understand wat are our need since we leave our home town... Hence, they organised a gathering dinner at the Banquet Restaurant in Kuching... In Kuching, Banquet Restaurant had been graded as 5 star restaurant... Quite expensive..

We are served with plenty of buffet style food... Got beef rempah, lemon fish, butter prawn, broccoli, fried chicken, mushroon soup, fried rice, fried bee hon, and many many more... Beside that, we are serve with variety of fruit too..

Mayang Tea - 1

On 14 & 15 October, Prof Kopli Bujang organized a field trip to Sarawak Tea Manufacturing Company- Mayang Tea Sdn Bhd. Mayang Tea Sdn Bhd is located about 90 km from the Kuching city. It take us about 2 hours drive from UNIMAS. This project was established on 1982 by Sarawak Land Consolidation and Rehabilitation Authority (SALCRA)...

This company is located between Kampung Mayang and Kampung Engkeroh.... It official website is http://www.mayang-tea.com.my/index_mayangtea.htm.

Mayang Tea is actually the lowland but not highland tea... So the tea is planted at an altitude of around 200 -800 feet above the sea level.. Lowland tea and highland tea different in taste and colour... For lowland tea, it has a very very strong dark brown in colour and strong taste... Mayang tea plantation consists of about 166 hectar areas o Native Customary Right (NCR) and it is owned by 174 Bidayuh families. Every year, Sarawak state government will subsidiary about RM 200,000 -RM 300, 000 to ensure the continuous operation and production of Mayang Tea Sdn Bhd.

We was brought to the plantation.. We are taught which part of tea leaves is the best to be processed... Normally the Mayang Tea plucker will pluck the first 2- 3 shoot of young tea leaves... After some explanation, we are allow to do our own tea plucking !!... Haha... it is really enjoyable...

This is how the plucker usually pluck tea leaves.. The first of 2 -3 shoots of young tea leaves.. According to the officer at Mayang Tea Sdn Bhd, processing the first 2-3 shoots of young leaves taste better...

Every morning, around 6.30am, the tea plucker will begin to pluck the tea leaves... Around 7.30am, they will finished plucking and sent the plucked tea leaves to factory to process.... They are kept inside the guni bag...

At the factory, the tea leaves are sent via the troller (red) in the withering room. The leaves is wilted for about 16 -18 hours...

Withering is done in troughs of 4.5' x 60' each which can hold about 900 kg of leaf.

After 16- 18 hours withering process, the tea leaves is then sent for rolling... In this process, the leaf cells are broken up... The release of the natural substances and flavor is regulated to give tea its aroma to develop after rolling...

The chemical components are mixed with enzymes which are released by cells in rotorvane machines via 3 passes. The rolled leaves are then separated into different sizes using rotary sifters.. After the separation, the leaves were placed onto Aluminium trays for fermenting in next stage. There are several machines which have been developed to achieves rolling process. But in Mayang Tea Plantation, the rotorvane is applied. Other types of machines like orthodox roller and C.T.C (Crushing, Tearing, Curling) machines or the combination of these machines are also used to perform rolling in others plantation.

Fermentation is the most fundamental process in tea manufacturing. In this process, different taste of tea can be produced by managing the fermentation time. In Mayang Tea Company, the crushed tea leaves are put into the tray and let it ferment for 2- 3 hours. They use the room temperature to carry out oxidation process and along this process the aroma is emitted. The fermentation process changes the leaves into different color and favor...

After rolling and fermenting, the tea leaves are transferred into drying machine.. The fermented tea leaves are put on the top of this dryer and were dried inside the machine. The temperature is at the range of 160 0F to 200 0. In this stage, the moisture of the leaves is reduced to 2 % - 3% and the fermentation process is stopped. After this process, the tea is used for grading...

This is the grading machine... Mayang Tea Sdn Bhd graded their tea into 4 different categories which DUST 1, DUST 2, DUST 3 and BOP.

The best category is DUST 3.... Normally, BOP is not used to make tea but manufactured to become fertilizer.

Kuching Festival

Every year in Kuching, there will be a festival known as Kuching festival where u could taste many different and special Sarawak Local Food.... This festival usually held on the end of Julai till the end of August - Duration for about 1 month... At that festival, u can eat alot of delicious food, unless ur pocket din bring enough cash... Haha

That place crowded with lots of people and lots of stalls... The price of the food is quite reasonable...

Gading Trips

In the mid of September, it is the world biggest Flower Rafflesia Glooming period... So, not to miss out the only opportunity, I followed my friends to Gunung Gading National Park to see such a wonderful and rare flower...

We went twice, the first time miss out the glooming period, so no matter wat we went there again for the second time just to see this flower... This flower only gloom once a year and every time glooming only last for 5 days....

Since all of us are already final year studied in UNIMAS, if not going to view such flower, then probably we don't have any more chances again... :- )

My friends are playing 'hide and seek' upon reach there on the first day...

Three of them is Playing Kung Fu.... Each show different technique of the Kung Fu... The first one (left) is playing ' Kung Fu Praying Mantis". The second one (center) Kung Fu is " Monkey style". The last one (right) is playing " Egret Kung Fu"... Each Kung Fu have its strength and unique martial art.

Sematan Palm Beach is full of jelly fish... Those who went there need to be very very careful... After u coming back from snorkelling, on the shores, we found plenty of jellyfish being traps on the beach and died... It is scary and frightened...

During the mid semester break on August, i and a group of friends went to Sematan Palm Beach for relaxing and snorkelling... It is a 2 days 1 night trip. This trips ruined me a lot... Not enjoy at all... : (

Having our free buffet dinner in the Palm Beach Resort Restaurant...

Every one of eat like an hungry ghost... Haha..

Each of us at least eat 3 round...

Before we coming back to unimas from Sematan Palm Beach, all of us take the opportunity to snap photo at the front entrance there... As a prove that we had been there..

We had our lunch at Ah Hock Seafood Restaurant... We just order seafood fried bihun and fried rice, the food serve to us is so special... Every one of us was in a shock when we are serve with those 'special food'.. All of us afraid that later our pocket will be in a big hole liao (Too expensive to eat it).. But its end up very very cheap only... Only cost RM 99 for both dishes serves with lots of seafood...

Proposal Presentation

2 September 2009....

This day is my most important day in this academic semester... It was my research proposal presentation.. On that day, my slot was actually arranged in the afternoon... But seem my supervisor was not free in the afternoon, she request the chairman person to squeeze in the morning session..

During presentation, i was so steady... Din feel any nervous... It might be probably due to enough practice. Besides that, i try to calm down my feeling by focusing my eyesight to just a audience only... By doing so, no fear will be feel...

But, for my proposal, i had make up a great mistaken and being questioned by one of the evaluator (Dr Hairul). He was actually questioning on my study whether there is necessarily to extract DNA from eggs white.. He told me that eggs white just contained only protein but not nucleic acid.... So it is useless for me to spend time looking for the DNA... Haha... Thank you to Dr Hairul for giving me good comments and advices..

忙碌了一个sem，终于有了休息的一天。。。。

因为今年是final year.... 所以非常的忙碌，忙着功课啦。。 FYP proposal 啦。。 FYP presentation。。堆积如山的assignment。。该死的考试。。。还有还要忙着进lab 做 research...

这一切终于来到了尾声。。。不过research 还是要做。。。得留下来继续做 research....

haiz... 真的好累！！好累！好想放一个很长很长的假期，好好的回家！！好想家！！好久没有回家了！！在外面这样久，总没有在家来得温暖。。。温馨的家。。

There are a total of 15 of us include all races... We have Indians and some bumi friends playing futsal with us.... They all are expert...

Official Visiting of TengKu Mahkota Johor To UNIMAS

This Monday ( 29/07/2009).... There is an official visiting by Tengku Mahkota Johor to UNIMAS around 2.30pm.... All the classes have been cancelled on that day..... Hooray....
(Becoz every lecturers need to go to give the warmest welcome to Yang Maha Mulia Tengku Mahkota Johor)..... So tua pai....

He is accompany by a group of policemen.... Plenty of Police superbikes parks at Rumah Universiti UNIMAS.

According one of my friend, she told me that Tengku Mahkota is riding a orange superbike... Tengku Mahkota wearing so usual.... Just wearing a branded T-shirt and Jeans....

This morning I was really really annoyed with the lousy system and poor serving of the librarian officer....

My laptop adaptor broke out recently and i have no choice to come on CAIS (library name) for completing my assignment and proposal... I need to use the desktop at the computer lab and open lab in CAIS... After i searching for about 15 mins, I cant even get a desktop to use.... Some don have electrical supply... Some cant access to the internet... Grr.... So frustrated....

Then, I go to approach the librarian at the counter... He is so lazy and he said later they will check... LATER!! How long they only wanna to check??? 15 min later?? 30 mins later?? 1 hrs later??? So sien with them... I have no choice... I contact my housemate, Steven to borrow laptop from him.... Luckily he is not using because he is having orientation activities with the juniors.... Oh... Thanks God...

But i still need to walk a long distance back to my hostel, unpacked and repacked my things and come to CAIS again.... The weather is damp hot .... Nearly become " roasted pig"...

Haiz... It is so troublesome by not using own laptop... ( Have own laptop but cant use it)... I hope that my new adapter will reach as soon as possible...

Yesterday morning, about 10am (not sure) Infinity FX at UNIMAS water towel is on fire.... The cause was believed came from the electrical short circuit... It was the overusage of electrical by the refrigerators... I am so pity for tauke... Luckily no one was get hurt upon this incident....

Hopefully, INFINITY FX will reopen soon.... I so miss their Mee Hong Kong... Their foods is extremely delicious...
I wan to eat mee Hong Kong from INFINITY FX!! Uncle, quickly reopen to do business again to cover up your loss from this incident....

世界音乐节 Rainforest World Musical Festival

2009 年7 月10日- 2009 年7 月12日，一连三天在砂拉越州举办了一年一度的世界音乐演奏会。。。是一项盛大的庆典。。。来自世界各地的音乐家都会聚集在文化村 ( Cultural Village), 大展拳脚。。。除了本土的音乐家外，还有来自智利（ Chile ), 加拿大/中国 ( Canada/China ), 匈牙利（ Hungary ), 法国( France ), 韩国 ( Korea )，美国 ( US )，芬兰 ( Finland)，纽西兰 ( New Zealand )，波兰 ( Poland )，葡萄牙 ( Portugal )，印尼 ( Indonesia )及坦扎尼亚 ( Tanzania )等国家。。。他们都呈现了他们国家的文化，音乐。。。非常非常韵人乐声！！

2009/7/10 - 2009/7/12， 3 days continuously at Sarawak Cultural Village held the Rainforest World Musical Festival.... It is an big occasion... There are Musician/ Performers came from different countries besides local musician...

虽然最近受到流感的侵袭，但是，此活动的热点不减，人潮很多。。。门票也很快就卖完了！！
Although, recently there was an outbreak of H1N1 in Sarawak, but this festival was crowded with full mountain of people.. None of them was afraid to get infected with H1N1 disease...

我也趁我最后一年在砂拉越就读，跟一群朋友去体验现场的气氛。。。

工业实习的经验之谈

从5月11 日至7月3日，整整两个假期都没有白白的浪费掉。这两个月时间说长不长，说短不短。在不知不觉中已经过去了。。。没有把这两个月大学假期白白浪费，比以前的假期过得非常有意义。。。哈哈！！:-} 至少，我是在学习不同的知识，不同的技巧，不同做人处事态度，而是沉迷在毫无目的的出街逛逛，看戏，见见旧朋友等等。。。

我在这时被大学安排到家乡（槟城）的闻名大马学府 ( 理工大学）进行工业实习。。 [ 其实，大学院方会帮我们寻找一系列的公司名单，让我们到那里去进行实习。如果我们对院方所找到公司不感兴趣，也可以选择自己所理想的公司]。寻找一间适合及属于自己的专修科的公司是非常非常的重要，因为它对你的未来真的很有帮助（如何去做你的FYP）。你们可以选择到政府机构如： Forensic Department, Forestry Center, Fisheries Center, Pusat Darah 实验所（Research Center）, 医院，大专，或是私人机构如： Path Lab, Stem cell company等等进行你们的工业实习。

在对的公司进行实习可以帮助提升你的实验里的技巧，应变能力，学习能力，及一些上司下属的沟通能力。。这些技巧，也许在课业上，你们是没有机会去接触到的。。。就打个比如，一些基本生物科技生该有的技巧如 Aseptic Technique, Pipetting Skills, Gel loading, Agarose gel preparing, Polymerase Chain Reaction (PCR)，机器的功能等等都是在课堂上读到的THEORY，但是没有机会进行practical。。。虽然，有时会有Lab Session, 但是， coursemate 如此的多，时间有限，根本没有机会让你接触到那些器材。。有时一些准备工作都已经由Lab Assistant 准备好了。。。

在这里想跟大家分享我一些经验。。。

我是在理大一位教授（Dr Alexander Chong）的实验室里向Master Students 学习。。这位教授的Field of Specification 是在 Aquatic。所以他的很多Research 都是跟鱼有关。他专研究zebrafish, ikan keli 和ikan haruan. 他出过很多很多的书及publications, 也得了很多奖。我被这位教授的才华感染了，我佩服他如此年轻就有了如此高的荣誉，我要向他看齐，希望将来也会向他这样有如此大的成就。。。当时，我被安排跟一位UNIMAS 毕业的Master Student 学习。这为Master Student 所研究的是Gene Expression on Zebrafish Using Hybridization Technique. (如果在行，应该明白是怎么样的一种研究)。我从他的身上学到了不少的技巧及知识。。。

初次报道，他先向我介绍及讲解实验室里的不同器材及运作，如Thermal cycler machine, gel electrophoresis, spectrophotometer, fume hood, laminar hood, microcentrifuge machine, microscope, oven, hybridization oven 等等一些基本的措施。。。在这两个月里，他也教我们如何prepare agarose gel, load gel, run gel, 运用pipette 的技巧， aseptic technique等等。。千万别小看这些基本知识，做起来可不简单，当中还有它的‘秘方’。。掌握不好这些技巧或不够熟练，可能会造成你得不到很好的实验的成果。当然，除了这些知识，我还得到了一些来自不同Master students 所分享的一些经验及劝告。。。除此之外，也认识到一些新朋友，真的投缘的是，我既然在那里遇到了一些失去联络的旧同学。。。（哈哈！！这个世界真的如此的微小，辗转几圈之后又重逢！！）非常非常怀念在那里的时光。。。非常享受在那里实习的日子。。。也非常想念我跟那群Master Students 疯狂的时候。。。

记得有一次，我跟三个Master students在Fish House 帮鱼交配后，无聊到跑到附近的大沟渠里捉泥鳅。。。在烈日下，我们真是太疯狂了，一个负责引秀泥鳅到另一段的沟渠，就那里我跟另外一个Master student 在那里站岗，他负责捉，我负责守。。。在我们三个人齐心协力下，成功捕捉到两条相当大泥鳅，大约有50cm 长吧!! 不错的收获！！让我深深感受乡间小孩童年时的喜悦！！也让我回味我童年的时光！！

另外，理大的实验室虽然没有砂大的实验室来得漂亮，拥有很大的活动空间，但是麻雀虽小，五藏齐全！！拥有尽有！！砂大远远不及理大。。。不愧是研究型大专！！实验室里的东西多到恐怖，有些冰橱都被逼放到实验室外的走廊里。。。每位Master Students 都没有自己的私人空间，私人座位或坐标，都是流动式的！！

好了，最后，我想忠告所有我的学弟学妹们，好好选择你们所理想公司，在工业实习时，不停的向人家请教，谦虚诚恳的态度去学习，或许，有些时候你已经懂了，但是并代表你已经掌握了，熟练了！！还有，经验告诉我，身为一位研究生，头脑必须清晰，了解你的实验的目的，需求，及程序！！一步一步有程序的安排你的实验工作。。。不要自乱阵脚。。要不断的提升自己，在大学三年所学是微不足道的，科技不断的更新，社会发展迅速，还有很多很多知识，大学教授是无法传授给大家的！

另外，也许在这时候，你们可以认真的考虑你们未来方向，想继续实验工作或从事商业！！当然，实验会给有些来说是件烦闷，枯燥的事情，肯定也不能向生意人这样赚取很多很多钱！！

如果想要继续做实验，可以选择了专读硕士，及博士学位，出来可以在大学当任教授或到私人公司去当位科学家。。。　或是，你也可以从事一些商业的活动，如做sales – 推销一些实验室必用品，化学药物，器材等等！！希望你们得到宝贵的经验！！

Farewell Party

Till the end of our internship, all the lab member is organized an farewell party for all of us...
Actually, this is also a farewell party for one of the international students, Mr Abdullah who have just pass his viva.
It is very enjoyable... All of them are so generous, offer themself by preparing delicious food...

All these delicious food are prepared by Mr Abdullah wife.... It is very tasty!! He told us that these are the food from his hometown (Yemen). Although some look similar to our food here, but when u taste it, it is totally different. Different taste, Different name and Different ingredient.

我在工业实习期间（Latihan Industri)，认识到一群很‘疯’ 的Master Students....

他们也许是吃到没有什么东西吃了，所以一时心血来潮跑到我们Fish House 附近的沟渠捉泥鳅。。。哈哈！！我也跟他们一起疯！！

非常不错的一个经验及回忆..... 好像我们童年在乡间嬉戏的时光！！很久很久已经没有这样疯了。。。

这是我们的收获！！一共捉到了2 只。。。好大的泥鳅（Ikan Keli)...

但是，他们下场会是如何呢？？

Enjoying Breeding Fish

This is 1 part of USM Fish House. Inside here where u able to find many aquatic lecturer research base. They rare the fish in this house. This corner is belongs to my supervisor, Dr Alexander Chong. Mostly of the fishes are zebrafishes, swordtail fishes, diskette fishes and etc.

One of our lab member is washing the aquarium. He is the graduated master student, going to continue his Phd at USM with Dr Alexander Chong too.

This small small fish tank where we used to breed pairs of zebrafishes. ( 1 male and 1 female is located at each tank). Then cover with dark clothes to create a period time in the dark.
Then, the next morning, once the dark clothes is opened, sunshine shine into the water. Once the divider is lifted up, pairs of zebrafish will start to mate and external fertilization occurs. Usually, after 20-30 mins, we stop them from further mating by separating them. Research have been to prove that the first 30 mins fish eggs is high in quality. So, we would only like to collect good quality of eggs.

Usually, all the master students will take turn to breed the fish, twice in a week in order to keep the stock. Normally, they will prepare 80 - 100 pairs zebrafish to breed.

Look!! I am watching the fish to mate and further collecting the fish eggs as a sample.

Usually, the male and female zebrafish will be separated by a transparents divider and kept them in dark period for 1 night. Next day, the divider will be lifted up to let the zebrafish to mate

See... ONCE the divider is lifted up, the male zebrafish is in a wild, keeping on chasing the female zebrafish. After a while, the female will release oocyte into the water while the male zebrafish release sperms. External fertilization occurs.

Woow... Plenty of eggs..... Their eggs are so tiny, soft and transparent in colour.

After 3 days, the fish eggs will hatch and growth into small tiny fishes

The First Day On Internship

I am having my internship in Universiti Sains Malaysia (USM).
11 May 2009 was the first day I start my internship. My supervisor is Dr Alexander Chong. He is a excellent and nice lecturer. Dr Alexander is specific on Zebrafish developmental stage and expression of particular genes. Very interesting !!
On that day, he asked me to meet him 11am at bio-school, USM.

That morning, i reach bio-school before the appointment time - around 11.00am. Since it was still early, i have a chance to take a walk to look around the surrounding and try to look for my supervisor room and lab. USM is not as big as i imagine. But if compare to UNIMAS, USM have more specification field than UNIMAS. I found parasitology lab, genomic lab, tissue culture lab, entomology lab, and etc.

Amongs those, that i found interesting is the entomology lab, inside the lab, there are research on Plasmodium sp. Mosquito. I saw the researchers, culture mosquito larvae, and adult mosquito too. I wonder how they do that. If accidentally released the adults mosquito from the containers, the whole lab will be contain with mosquito that spread malaria and all the lab member will be infected.

After i walk around, i went to visit my other coursemates working lab. Mostly, they have nothing to do since the whole morning. Their supervisor just asked them to read a lot of journal.
Around 12.30 pm, my supervisor is free, he managed to meet already. ( Haha!! He is such a busy guy, only available to meet after 1 hours from the appointment )

He brought me to his office. Wah, His office is very messy, full with aquarium. Later, he went into the lab to introduced me to 1 of the Master Student whom was also graduate from UNIMAS bResource Biotechnology too. His name is Kah Loon, my direct senior... Kah Loon now is doing research on the gene expression of Iclp 1 and Iclp2 into zebrafish whole-mount in situ hybridization. At first, he introduced me to all the member of the lab. ( Gosh!! The world is really so small, i met 2 of my secondary old friends there.)

After this, we went to lunch together with others master and Phd students. After coming back from the lunch, Kah Loon briefed me about functions and usability of instrumental in the lab. They have different kinds of freezer, gel electrophoresis, spectrophotometer, trans-illuminator, incubator, water bath, In-situ Hybridization Incubator, thermol-cycler,autocave machine, shaker, and etc.

Although the lab is completed with all basic instrumental, but there is a bit of space constrain. Too pack already, even done have enough working place, all the Master Student also done have their own personal place. Even at the Corridor there, also full with machine. Blocking the path....
That is the only things that worse than UNIMAS. Air -cond also not cool enough. Sometimes, power supply also lacking.

青山岩

青山岩非常的靠近Kampung Bako, 从那里到青山岩只需要5 -10 分钟路途。。。是一间佛寺来的。。。风景很美。。我们那天在到码头做船到Bako 之前，就去了这个佛寺求个平安。。

哈哈！！看我变身！！千手观音上身咯！！

Bako 之旅

这旅程真的是非常非常爽，非常非常的开心。。。不知道为什么，也许刚考试了，什么压力都没有了，大家都可以尽情的享受大自然。。。

Taman Negara Bako是一个在Kuching, Sarawak 历史悠久的国家森林保护区。里面常有很多很多各式各类flora and fauna。。其中可以看到最稀奇，罕见的动物有：长臂猴，有胡须的野猪，松鼠，猴子，四脚蛇，各种各样的草本植物。如果喜欢大自然生态的人就会非常的开心。。。

凌晨5点多左右，可以看看漂亮的日出，听听小鸟的鸣叫声，吱吱喳喳的，非常的悦耳，动听！！晚间，可以听见不同的昆虫的叫声。。听听海浪波涛汹涌的在怒吼。。也可以吹吹清凉的海风。。。

去Bako National Park必须乘坐一艘中型的motor boat 从 Kampung Bako 出发。旅程大约20 - 30 mins.

我跟我的女朋友在乘船的时候拍的。。。。嘻嘻，被海风吹得一头散发。。当时我们在烈日之下，搭了一艘没顶的船。。。晒到挣不开眼睛。。。

喔！！终于到了！！ Bako National Park 我们来也！！

你们有看过一只实魂的野猪在海边散布吗？？在Bako 你们就可以看到！！这只野猪好像失恋了，在海边散心！！野猪啊，你可千万别想不开啊！
哎呀，天涯无处无芳草！！

我，我的女友及她的朋友在沼泽林附近合照。。。我们都准备去爬山了。。。

沼泽林！！海水涨的时候！！

我们爬山爬到一半，忽然下起雨来！！大家都穿着雨衣爬着下来！！回到宿舍，大家的衣服，裤子，鞋子都湿了！！

结果，大家都忙着把湿湿的雨衣在梁上晒！！在一旁，也在晒鞋子！！

这是我们晚间跟着一团人去jungle trekking！！导游发现一只有毒的青蛇在树上！！据说，这只蛇都是晚间才活动，都有很强的thermal receptor。。。就是利用这个receptor 来扑捉猎物，如：鸟类！！

小强！！你这么出现在这里？？？

哇！！好大只的竹节虫！！

好大的一只蜘蛛！！有毒的！！各位，如果你们被咬，一定变成蜘蛛人了！！哈哈！！

母亲节

每年5月的第2个星期是母亲节。在这一天，相信每个孝子孝女都忙为这一天的来临而跟母亲一起度过。。。在这一天，你如何和母亲一起度过呢？？如何表达你的孝心呢？？

送给母亲最温馨的拥抱？？一起吃大餐？？送康乃馨给母亲？？买些礼物送给母亲？？
嘻嘻！！以上种种的我都没有做到？？

在这一天，我不知道该如何表示，不知道该如何表达我的孝心，也不知道表达我的歉意。。。因为我实在伤透了母亲的心，一次又一次的让她伤心难过。
一次又一次与她顶撞。。她不喜欢我这么做，但我扁扁不听，持续这样做。。。
她的叮咛，我却认为是唠叨，整天喋喋不休说个没完没了。。。

虽然，我买了一罐护肤品，可是任然在母亲节那天，没有拿出来送给她。。。
因，我怕她不喜欢。。。
虽然，母亲节当天，母亲收到了很多很多朋友的讯息祝福，
可，我一封讯息或祝福都没有送给她。。。
虽然，母亲节当天，母亲常挂在嘴边，暗示着，
但，我还是无动于衷。。。

咳。。。我真是一个不孝的儿子。。。没有进孩子的责任。。。一而再，再而三的骗她，伤她的心。。甚至一声慰问我都没有。。。

我对此感到非常的惭愧。。。
妈妈，我错了！！原谅我这个不孝儿！！

可是。。。。

在外地读书，兜兜转转已经半年过去了。。学期考试考完，就要回家了。可是，我一点都不想要回家。。。心情非常的纳闷。。。情绪非常低落。。。没人能够与分享我心中哀伤。。

那时我在想。。。

“ 我想到一个世外桃源、与世隔离的地方。。把所有的烦恼，所有的悲伤，所有的过错，所有的压力，所有的一切一切都留在这里，而自己就逍遥的躲到那个地方。。。 ”

为何我会有这个感觉？？这种感觉是前所未有的。。。

我的家到底发生了什么事？？

为何我回到一个我却感觉不到温馨的家。。。

顿时开始觉得我对我这个家已经渐行渐远。。。

顿时开始察觉到我跟家人的代沟越来越深。。。

我开始不了解家人，家人也开始不了解我。。。

家人开始对我不信任。。。（我讲了种种的骗话所致）

常常与家人在有些意见上起了冲突，三两天就争吵，情况搞到很僵。。。弄到大家的心里都很难受，都很不安。。。

我实在伤透了父母的心，没脸回家去面对他们。。。不知道要如何面对。。

不过，那个家始终都是要回的。。。我始终都必须回到原来的地方 - 一个我成长的地方。。。

如果我继续在逃避，问题依然存在，反而与家人之间的出现的裂衡会越来越深。

所以我要勇于面对，要尝试去补救我犯下的祸根，以求得他们的原谅。。。不然，到时后悔就来不及了

这一天真的是很难忘的一天。。。。（其实不是什么大日子，只是发生一些事情）

这天，我，我的女朋友及她的姐妹桃们就相约一起去出去吃火锅。。。大家都吃得相当的开心。。。大约11点就回到宿舍了。。。可是一时的贪玩，就打算在去游车河。。。

结果，大家就一同出去了。。。可是固执的我，就坚持不要打油。。一直都有这个想法，整桶RM 2 的油在油箱里是足够的。。。足够让我们驾整整200km 的来回路程（哈哈！！未免太天真了吧）。。。

事与愿违，我们的车子驾到 kuching civic centre 附近就没有油（在斜坡中停下）。。而且那边是位于公路中，车子来的速度非常的快，非常的危险！！非常恐怖！！

在这样的情形，我们只好打电话找朋友求救。。只希望能够找一个朋友还没有睡觉的（当时是半夜11 点55 分左右了）。。还好，打了几通电话，终于给我们找到了一个朋友的帮忙。。。（真的是很嗅！！）

我们整辆车子中只有我一位男生，其余的三位都是女生。。我们就傻傻的在车上等待着我们的救星的到来。。。因为当时的地点是相当的暗，大家都把门锁上，对着时间倒数！！

半个小时后，我们的救星终于来了。。成功的帮了我们脱险。。。我们大家大约1点也安全的到宿舍了。。。（在回家的途中，大家都有阴影，所谓一日被蛇咬，十年怕草绳）！！

从这次的教训中，我以后不敢在固执了。。这次真的是时时刻刻牢牢烙印在我的记忆中。。。

今晚宿舍的wireless 非常的好。。。到这样夜都能够上网，真的很不错。。。

我这个傻子，有觉没得睡，一个独自对在电脑的荧幕发呆。。我怎么会搞到这样迟了还不睡呢？？到底做什么东西喔？？哈哈！！

其实我是有点倦了，可是没有办法， experiment report 没有写好，明天要交了。。所以就要开夜车咯。。。哈哈。。这就是大学生活，这就是所谓的大学生的气质，不到最后一分钟，功课是不可能去做的。。。

到底我们的现任首相，拿督斯里納吉能够就任多久？？大马政坛上又会有了什么变化？？什么把戏？？哈哈！！

不知道我们的拿督斯里安华是不是又会再次被判 ‘鸡奸’ 罪呢？？

何谓大学生活？？

最近，　本人顿时有了一些的感触。。。　
本人在想，　要如何让自己的大学生活过得灿烂？？　
要如何去充实自己？？
怎样让自己的大学生活过得有意义呢？
（因课业的繁忙，无法找到平衡点，开始感觉大学生活过得如此的烦味）- （好没有意思啊）。。。
开始厌倦了这样的生活。。。。咳。。。

有时，这些问题会不断不断的出现在脑海中
为何有些人可以相当悠闲？？　（还有时间看戏，打game, 拍拖等等）　
为何有些人又显得繁忙？？
为何有些人能够在繁重的业务中，　过得悠闲且轻松的生活呢？？　
而有些则不能呢？？
甚至有人能够兼顾学业，有能活跃在大学生涯中的种种活动。。。
（而我为何不能呢？）

他们的秘诀在那里呢？？
是不是他们读的书不一样？？读的课程比较轻松？？是不是他们有过人的先知？？还是他们拥有比我们大家多出的24 小时可用？？

其实：秘诀是在如何去安排好他们的时间。。。
在大学生活里，我认为时间的规划是非常非常的重要。。。
时间安排的好，做起事来也非常顺利，也会比较起劲。。。自然而燃的，过的生活也会比较悠闲，没有什么压力。。。

我看到大部分学生的一个坏习惯，就是;
凡是不到最后一分钟，就是特别没有所谓的 '灵感' 来完成你手头上的课业。。一天拖一天，结果呢，课业就越堆越多。。直到最后完全崩溃。。。开夜车。。。赶通宵。。。做出一份底水准的功课。。。第2天早上，各个就变成了睡星公。。。各个都有了一个很深的熊猫眼。。。。
（其实本人也是属于这个族群的- 哈哈哈）。。。

时间是在你不知不觉中一滴一滴流走。。。。朋友们，清醒吧！！赶紧把握住你的时间，一分一秒的守候它，不要虚度光阴。。。

Yesterday (20 March 2009)
Task : Extraction the DNA from the liver tissue.

Before we start our experiment, our lecture is giving some briefing regarding some precautions and methods to take....

Before that, scalpel blade is 'install'...

Then the liver cell is minced into small pieces....

After incubation for 1 hours, Phenol:Chloroform is added and it must be done in the Laminar flow hood....

Then, centrifuge the sample and u will obtain the results as below....
Transfer the supernatant (aqueous phase) into new sterile microcentrifuge tube...

Everyone of is busy perform their own experiments...

Yesterday, It was raining heavily. While waiting for the bus, i and my "gal"friend play bowling at our Unimas bowling center. There are 4 of us playing this game.... HAHA!!! Done of us are expert... WE ARE BOWLING NOBE!!

Statistic on visiting CAIS

CAIS stand for Centre for Academic and Information System
It is UNIMAS library- a place where UNIMAS students spend most in there to do revision, doing assignment, looking for book and etc....

There are plenty of facilities such as personal carrel room, discussion room, 24 study room, online area and air-condition.... It is very convenient and comfortable for students to do their revision.

Below are survey that are done by CAIS adminitration:

Figure 1 show that are 40% of students feel that CAIS is an important place for them to study and doing research

Since almost 70 % of students think that CAIS located at a strategic place and very convenient for them to come to CAIS but so sad to see that only about 15% of students always visit CAIS....
From here, it can prove that Malaysian university students done fully utilise the facilities provided....
Wah!!! Almost Half of students in UNIMAS never used discussion room and even personal carrel room...
This statistic have proved my saying above: Students do not know how to fully utilise the facility provided.... Haizzz....

The operation hours of CAIS is from 8.00a.m till 11p.m during weekdays while during weekends, the operation hours is from 9.00a.m till 10.30 p.m.

So, 24 hours study room is open 24 hours for students to study till mid night if their hostel cant provided a silent and quiet place

Plant Genomic DNA Extraction
Last week friday, we was given a chance on running an experiment- Plant Genomic DNA extraction.... It is very interesting..... and gain a lot of knowledge and skills

Plant Acacia mangium had been used on this experiment. The experiment started by doing some cleaning on the Acacia leaf then followed cut it into small pieces.... ( This methods make it easy for disrupting the cell wall of the leaves)

Later, Liquid Nitrogen is added into the mortal in order to freeze the leaves... then the leaves is rapidly grind into a fine powder with a pestle and mortar as the liquids nitrogen boils off (Liquids nitrogen is added to keep the powder from thawing while grinding)
* Note: Liquids Nitrogen is extremely harmful and extra precaution must be taken
Remember to wear double layer of Gloves...

In order to prevent contamination, all of our experiment is done on the laminar flow hood... Due to laminar flow hood is not enough to provide for all 139 of biotechnology students, so we have to share each others.. Queue up in a line...

This is the sample after CIA solution is added and centrifuge at 13000rpm for 5 minutes to obtain supernatant. The supernatant is carefully transfered into the sterile, new, clean 2.0ml microcentrifuge tube

This is the sample after cold isopropanol is added and centrifuge at 13000 rpm for 2 minutes in order to precipitate nucleic acids. Then after that it is store at -20 degree celcius freezer for at least 30 minutes.

Plenty sample have been prepared.

Look!! One of friend is pipetting the supernatant out from the centrifuge tube into a new tubes...

After 30 minutes, centrifugation at 13000 rpm for 2 minutes to obtain the pellet DNA. After the pellet of DNA is obtained, the supernatant is discarded. Then DNA wash buffer is added and rinse the pellet by tapping the tube gently with finger tips ( the dilute ethanol removes salts ).
After that, centrifuge again at 13000rpm for 2 minutes. Supernatant is discarded and let the pellet air dry before resuspend in TE buffer for further analysis.

Lab Experiment ( DNA Isolation - Methodology)

Methodology:

A. Preparation of Overnight Bacterial Culture

1. A single colony is inoculated by using autoclaved tips from the agar plate containing single colonies of Bacillus amyloliquefaciens UMAS 1002.

2. The tip with Bacillus amyloliquefaciens UMAS 1002 is added into bijou bottle containing 5 ml of LB media. The culture is then incubated with shaking for overnight at 370C.

3. The bacterial cultures are kept at 40C for used in further DNA analysis and isolation.

B. Isolation and Preparation of Genomic DNA from Bacteria

1. One and a half of an overnight culture is transferred into Eppendorf tube and centrifuged for 30 seconds.

2. After centrifugation, the supernatant is removed and the cell pellet is resuspended in 567µL TE buffer. The cell pellet is mixed well by continuous pippeting.

3. 30µL of 10% (w/v) SDS and 3µL 20mg/ml Proteinase K is added to give a final concentration of 100µg/ml proteinase K in 0.5% SDS solution. 100 µL of 5M NaCl solutions is also added and mixed well.

4. 80µL of CTAB/NaCl is added into the mixture. The culture is mixed well and incubated for 10 minutes in water bath at 65¬0C.

5. After 10 minutes, 780µl of Phenol / Chloroform / Isoamyl alcohol (25: 24: 1) that is added to the mixture. Then, the mixture is vortexed briefly and centrifuged for 5 minutes to separate the phases.

6. The viscous and clear supernatant is transferred carefully into new Eppendorf tube. The aqueous DNA layer once again is re-extract using Chloroform/Isoamyl alcohol (24: 1) and centrifuged for 5 minutes.

7. After centrifugation, the supernatant is transferred into a new Eppendorf tube. 420µl of isopropanol is added to precipitate out the nucleic acid. The tube is vortexed up and down slowly until the white DNA precipitate appears. The DNA precipitate can be pelleted by centrifugation for 30 seconds.

8. The supernatant is removed and the DNA pellet is washed with 200µL of 70% ethanol and centrifuged for another 30 seconds at room temperature. The supernatant is carefully removed and the pellet is air-dry.

9. The dried DNA pellet is dissolved in 100µL of TE buffer and stored at -40C before use in the further analysis.

C. Isolation of Double-Stranded Plasmid DNA

1. The bacterial cells are harvested from a 2 ml overnight culture containing cloned plasmid by transferring into a 2 ml Eppendorf tube and centrifuging at 8000 rpm for 2 minutes at room temperature.

2. The supernatant (culture media) is removed carefully and recentrifuge the pellet for 1 minute. Any traces of liquid media are removed completely from the tube.

3. The cell pellet is resuspended by vortexing briefly for 10 seconds using 100µl of Solution I. and after that kept the tube on ice.

4. 100µl of Solution II is added to the cell suspension and gently mixed by inverting the tube 10X. The tube is left at room temperature and the lysis reaction is allowed to occur for 5 minutes. A clear viscous liquid have been observed.

5. 300µl of Solution III is added and mixed by inverting the tube 10X. A white precipitate has been observed. The precipitate is pellet out by centrifuging at 10,000rpm for 5 minutes. The supernatant which contained plasmid DNA is transferred carefully into a sterile 1.5ml Eppendorf tube.

6. The plasmid DNA is precipitated by adding 800µl of cold absolute ethanol. The content is mixed gently by inverting the tube at least 10X.

7. The plasmid DNA is pellet out by centrifuging at 13000rpm for 5 minutes at room temperature. The supernatant is discarded and washed the pellet with 500µl of 70% ethanol and recentrifuged at 13000rpm for 2 minutes.

8. The supernatant is discarded as much as possible and allowed the plasmid DNA pellet to air dry for 15 minutes at room temperature. The plasmid DNA pellet is resuspended and dissolved in 50 µl of sterile ultra pure water.

D. Restriction Analysis of DNA Using Universal Restriction Endonucleases.

1. The reaction for restriction digestion is prepared as the list order in the following:

Sterile ddH2O 12µl (quantity sufficient)
10 X assay buffer 2µl (one-tenth volume)
DNA (with insert) 5µl
Restriction enzyme (Pst 1) 1µl (1-10 units per µg DNA)
Total volume 20µl

2. After finished adding the above materials, we gently mixed the digestion by pipetting and incubating the reaction at the 370C for 1hours – 2 hours.

3. In the mean time, the isolated genomic DNA and plasmid DNA are ran through by using Agarose Gel Electrophoresis method 1% of agarose gel is used and electrophoresis is performed at 105 volts for 30 minutes.

4. After 2 hours, the enzyme is inactivated by heating at 650C for 10 minutes.

5. Then, the restriction digestion sample is ran through Agarose Gel Electrophoresis methods in order to get the DNA band.

E. Glass Wool Spin Column Preparation.

1. A clean hypodermic needle is used to drive a hole in the end of a 650 microliter Eppendorf tubes from the inside and then the cap is cut off.

2. Gloves are worn in order to pull off a small piece of glass wool and roll it into a ball between hands. The glass wool is place in the bottom of the 650 microliter tube.1 ml pipette tip is used to pack the glass wool in the bottom of the tube.

3. The tube with the glass wool is placed into a 1.7 ml Eppendorf tube.

F. Eluting DNA from Agarose Gel Fragments.

1. Ethidium bromide stained agarose gel is visualized with a transilluminator on low setting to get the fragment of interest. Then the fragment of interest is excised out with a clean razor blade in a fast pace in order to minimize the UV exposure to the DNA. After the excess liquid is removed with a tissue paper, the agarose fragment is placed into the spin column.

2. The tube at 2874 Xg is centrifuged for no more than 45 seconds to elute the DNA.

3. Transilluminator is used to check the eluent in the bottom of outer tube for the presence of ethidium bromide stained DNA.

Lab Experiment ( DNA Isolation - Intro)

Isolation of Genomic DNA and Double-Stranded Plasmid DNA from Bacterial Culture with Restriction Analysis of Genomic DNA Using Universal Restriction Enzymes and DNA Extraction and Purification.

Objective:

1. To use the aseptic technique in transferring bacterial colonies to bacterial growth media.

2. To learn the technique of vortexing or pipetting in order to mix well the solutions.

3. To learn technique on how to isolate the genomic and plasmid DNA of the bacterial culture.

4. To do the Restriction Analysis using Universal Restriction Enzyme

5. To learn the technique of DNA extraction and purification from Agarose Cell.

6. To learn the technique in performing the Agarose Gel Electrophoresis.

7. To learn the skill of separating the supernatant from the pellet.

Introduction:

Genomic DNA is the DNA found in the chromosomes. It can be either double-stranded or single-stranded. In modern molecular biology laboratories, DNA is routinely isolated from cells as the first step in analysing genes.

In order to obtain the purified DNA from cells, scientists must first release the DNA by breaking open the cell membrane and the nuclear membrane. Once the DNA is released from the nucleus, however, it can be destroyed by enzymes in the cytoplasm that degrade DNA. As a result, there are some ways to guard the DNA against attack by enzymes from the cell. Lastly, the released DNA must be separated from the proteins, lipids, and carbohydrates, amino acids, fatty acids, and other cellular molecules.

There are two features of most DNA isolations that help protect the DNA. The first is coldness which is isolation of DNA procedure is usually done on ice to slow the action of cellular enzymes that might degrade the DNA. The second is gentleness: DNA strands are extremely long and fragile. Therefore all steps should be done slowly and gently to avoid shearing the DNA strands. The same things go for the plasmid DNA.

There are some methods to extract the plasmid DNA that is either using the spin column or the conventional methods. As we know that, bacterial cell contain both the genomic DNA and plasmid DNA, scientists need to isolate the plasmid from the genomic DNA. The most suitable procedure is used to extract plasmid DNA from bacterial cell suspensions based on the alkaline lyses procedure developed by Birnboim and Doly. Both nucleic acids and proteins denature under the alkaline conditions, and once the solution is neutralized by the addition of Potassium Acetate Salts, chromosomal DNA and proteins aggregate together while the plasmids renature correctly and remains in the solutions. This procedure take advantage of the fact that plasmids are relatively small supercoiled DNA molecules and bacterial chromosomal DNA is much larger and less supercoiled. As a results, the chromosomal DNA unable to renature back correctly.

Restriction Enzyme (or restriction endonuclease) recognizes a specific base pair sequence in DNA called a restriction site and cleaves the DNA within or near that sequence. All restriction enzymes cut DNA between the 3’ carbon and the phosphate moiety of the phosphodiester bond. It can be either in the form of sticky end or blunt end.

It is encouraged to use more than 1 restriction enzyme in the restriction digestion if both enzymes are active in the same buffer and the same incubation temperature. But in this experiment, we are just using only 1 restriction enzyme that is Pst 1. Small amount restriction enzymes are needed in the digestion solution depending on size of the DNA being digested. Larger DNAs should be digested in larger total volumes (between 50 -100µl) and vice versa. Besides that, different enzymes require different buffer solutions. Enzymes will only perform effectively in appropriate pH, salt concentration and temperature. In order to choose the appropriate assay buffer, we need to refer to the vendor’s catalog for the chart of enzyme activity.

Agarose gel electrophoresis is an easy method to separate DNA fragments by their sizes and visualize them. It is a common diagnostic procedure used in molecular biological labs. The technique of electrophoresis is based on the fact that DNA is negatively charged at neutral pH due to its phosphate backbone. For this reason, when an electrical potential is placed on the DNA it will move toward the positive pole. The plasmid was found in many different supercoiled forms in the bacteria. When plasmid was isolated from a bacterial culture, all the different supercoiled forms of the plasmid was isolated, and each will migrate differently on the gel, producing about three major bands and many minor bands. When this mixture of supercoiled plasmids is cut with a restriction enzyme, the different forms linearize and unwind. As a result they all become identical and run at the same rate, and usually only one band seen on the gel.

Elution is a method of quickly purifying agarose gel DNA fragments for use in subsequent reactions such as further restriction enzyme modification, T4 ligase, Taq polymerase, Klenow or polynucleotide kinase. Although higher yields of purified DNA can be obtained from commercially available purification kits, greater ligation efficiencies per given amount of DNA have been seen with the use of spin column.

我看到我的一位朋友尽然在课堂上睡觉，而且还是睡到很熟。。。。
I saw 1 of my friend was sleeping while lecturer was given lectures on the hall....

不知道昨天晚上做了什么。。。哈哈。。。一定是去‘偷鸡’，所以睡眠才不够。。这一幕就这么巧给我捉到正着。。。

其实，早在上个礼拜开始，我们实在是很忙，忙到连睡觉的时间都没有。。。太多太多的Assignment 及考试。。。所以呢，我的这个朋友实在是顶不过睡神的诱惑，就盛这个讲师不注意时，这样的一溜，就睡到桌底里去了。。。。

最近，assignment 一大堆。。。考试一科接一科的降临。。。

这真的另到我的头很大。。。忙着安排好我的时间。。。每科科目的考试时间及assignment 的截至日期都一一记了下来。。。

Recently, my got full mountain of assignment coming non stop... After finished settle and pass up 1 assignment, come again 2-3 new assignment title... Its MAKE CANT　BREATH.....

In order not to miss out, I try to manage to manage my time.. Write down the due date of the assignment and the exam period.

看！！妈的！！二月及三月的时间都相当的满。。。似乎每天都排到满满！！
朋友们啊。。。试问你们在你们的大学的生涯里，都是这样的忙吗？？

See.... My February and March Schedule is Almost Full. Haiz... This is the university life....
I done know how are my friend time schedule in others university.... Is their university life also so compact???
